Her most famous characters on The Simpsons are Agnes Skinner, Lindsey Naegle and Brandine Spuckler. Her performance as Mom is her most noteworthy Futurama role. In the Club Penguin Disney TV Series, she was the voice of the Penguins as well and the voice of Daisy Duck.

Before becoming a regular on an animated TV show, MacNeille worked in a variety of jobs and had numerous minor voiceover roles. For ten years, she was also a member of The Groundlings improvisational comedy group. MacNeille took acting workshops and worked as a casting assistant for voice acting talent agent Bob Lloyd. Lloyd and fellow agent Rita Vennari got MacNeille her first role in an animated show: a part in an episode of Scooby-Doo 1979 and Scrappy-Doo.
She sang and appeared in the music video (as Lucille Ball) for the song “Weird Al” Yankovic’s song “Rick” in 1973. The song was based on the I Love Lucy television show and parodied Toni Basil’s song “Mickey”. MacNeille also appeared on the tracks “Pretty Fly for a Rabbi” and “Jerry Springer” on Yankovic’s 1999 album Running with Scissors.
MacNeille was cast as Babs Bunny in Tiny Toon Adventures from 1990 to 1995. Writer Paul Dini said that she was good for the role in Tiny Toon Adventures. Since she could do both the voice of Babs and the voices of her impressions. The success of Tiny Toon Adventures led to the series of Animaniacs. MacNeille was brought in to voice Dot Warner which is one of the three main characters of the show. Because character of Dot’s was very similar to Babs Bunny. Andrea Romano, voice director and caster of Animaniacs, said the casters had no trouble choosing Dot’s position. MacNeille was nominated for an Annie Award for her performance on the show in 1995. She has provided voices for numerous television shows, films, video games and commercials. She has earned over 200 credits.
She has been the voice of Daisy Duck and Wilma Flintstone since 1999 and 2000 respectively. In Elvira, Mistress of the Dark, MacNeille also appeared as an angry anchorwoman. She also served as the voice of the great-aunt of Elvira, Morganna Talbot. For the 2003 Wile E. Coyote and the Road Runner short feature, The Whizzard of Ow, she provided voice acting. MacNeille has provided voices such as Chip’n Dale Rescue Rangers (as Chip and Gadget), Histeria, Rugrats (as Charlotte Pickles), Hey Arnold and many other cartoons and TV Shows. She also has done dubbing work on anime translations in English.
